Une fois de plus, Skipper, Kowalski, Rico et Private, dictent leur loi à l'intérieur du zoo de Central Park. Afin de pouvoir garder leur voisinage heureux, les pingouins doivent parfois s'aventurer dans la jungle new-yorkaise afin de ramener de nouvelles trouvailles qui pourront les aider à préserver leur tranquillité. Mais alors que les quatre frères pensent avoir un contrôle absolu sur les autres animaux, un nouvel arrivant fait son apparition : Julien, le roi des lémuriens, doté d'un égo surdimensionné, il passe ses nuits à faire la fête avec ses copains. Exaspéré, les pingouins vont devoir rivaliser de nouveaux stratagèmes pour récupérer leur territoire.
