Nul ne sait qu'au-delà de la Terre, s'érige le puissant royaume d'Asgard, patrie des Dieux scandinaves sur lesquels règne le tout-puissant Odin d'une main de fer. Sur Terre, Thor, dieu de la foudre et fils d'Odin, fait régner l'ordre et la justice à l'aide de ses fantastiques pouvoirs et surtout, son marteau Mjolnir que lui seul peut porter par la seule force de sa grandeur d'âme. Mais Thor a en fait un secret : alors trop prétentieux et orgueilleux, il fut banni d'Asgard il y a longtemps par son propre père et fut condamné à partager son corps avec celui d'un humain, afin qu'il apprenne l'humilité. Ainsi, il cache désormais son identité sous les traits du timide docteur Donald Blake, un infirme. Lorsque le danger se fait sentir, Blake frappe sa canne sur le sol et devient ainsi Thor pour combattre le mal, mais s'il perd son marteau, le charme se rompt et Thor reprendra sa forme humaine... Mais Odin n'avait pas prévu que Thor tomberait amoureux d'une mortelle, Jane Foster, l'infirmière qui seconde Donald Blake et qui ignore tout de sa double vie ! Pour le maître d'Asgard, têtu et fier, un Dieu immortel ne peut épouser une mortelle, et ainsi le père et le fils s'opposent-ils souvent au grand désarroi du souverain. Hésitant à révéler son secret à sa belle, au risque de provoquer la plus grande colère d'Odin, Thor doit aussi affronter des ennemis réguliers, tantôt des supervilains de la Terre aux pouvoirs fantastiques comme Mr Hyde et le Cobra, mais aussi des créatures divines et autres créations de ses ennemis, à l'instar de l'enchanteresse, l'éxécuteur ou l'homme absorbant. Toutefois, son plus mortel ennemi n'est autre que Loki, le dieu du mensonge et du mal, le propre demi-frère de Thor, jaloux de ce dernier. Loki est sans cesse derrière les plus viles machinations et fera tout pour discréditer Thor aux yeux de son père pour s'assurer un jour la domination suprême d'Asgard...
