Au cours d'un voyage scientifique dans l'espace, 4 amis, Reed Richards, sa femme Sue, Johnny le jeune frère de celle-ci et Ben Grimm, sont pris dans un orage cosmique qui a d'étonnantes retombées sur leur organisme... Ils s'aperçoivent en effet vite que les rayons cosmiques les ont pourvus de capacités exceptionnelles : Reed peut s'étirer à volonté et transformer son corps en caoutchouc, Sue peut se rendre invisible et créer des champs de force, Johnny peut s'enflammer des pieds à la tête et s'envoler... tandis que Ben se mue en une Chose, vrai colosse de pierre, mais sans pouvoir reprendre son apparence normale...
